5-Amino-1MQ (also known as 5-amino-1-methylquinolinium; CAS 42464-96-0) is a synthetic methylquinolinium derivative that functions as a highly cell-permeable and selective inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT).
Mechanism of Action
NNMT catalyzes the methylation of nicotinamide (a form of vitamin B3) using S-adenosylmethionine (SAM) as the methyl donor, producing 1-methyl nicotinamide and consuming methyl groups. By inhibiting NNMT, 5-Amino-1MQ helps preserve nicotinamide and related NAD⁺ precursors, thereby enhancing intracellular NAD⁺ availability. Elevated NAD⁺ supports mitochondrial oxidative metabolism, sirtuin activity, and other NAD⁺-dependent processes.
Preclinical Findings
- In diet-induced obese (DIO) mice treated with 5-Amino-1MQ (20 mg/kg three times daily, subcutaneously, for 11 days), treated animals lost approximately 5 % of body weight relative to baseline and significantly reduced adipose tissue mass and adipocyte size without any change in food intake.
- Adipose reductions were around 35 % for epididymal white adipose mass, with adipocyte size and plasma cholesterol lowered by roughly 30–40 %.
- In vitro, 5-Amino-1MQ shows strong cell permeability and high selectivity for NNMT, with minimal inhibition of other methyltransferases at relevant concentrations.
- Additional studies combining NNMT inhibition with dietary restriction suggest additive improvements in metabolic efficiency.

In aged mice, 5-Amino-1MQ has been associated with improved muscle strength and faster recovery when combined with exercise, possibly via NAD⁺ preservation and improved mitochondrial health.
